What do you mean by "block up"?
You can seal off a fireplace and even build a wall over it (that can be removed later), if you wish.
To seal off a fireplace, just close the damper and stuff an insulation batt up into the top of the firebox against the damper.
Put a piece of sheet metal held down with a cement cap block or bricks over the flue opening on top of the chimney to prevent rain and snow from getting down into the flue.
It can easily be reopened later.
Good Luck!
Mike
