If I have a 16x16 room, do I install the OSB T&G subfloor parallel to the floor joists? What exactly does the T&G do?

You will want to lay it long ways across your joists. The T&G gives support between the joists, keeps monsters from crawling up into your room....naw, that ain't right. My grand daughter is helping me.
Really, run it across the joists for proper support.

Don't forget the construction adhesive, a 3/8 bead on every joist, double bead on the joints, and an 1/8" bead in the groove. Let the sheathing down, don't drop it on the joists. Start nailing on the toungue side, then middle, working out to the ends each way. Leave an 1/8" gap at end joints for expansion. Stay 2" away from grooves when nailing, use ring or better yet, screw shank nails, or screws. Pull all air (miss) nails. If possible, check for shiners (air) underneath, remove and renail them. No squeeks.
