I have a microfiber couch that has been in a barn in storage.

It has some mold underneath and mice seem to have traveled across but have not damaged it (ie chewed it or anything) but there is poop and urine on it.

What is the best way to clean it. With a steam cleaner or cleaning solution and elbow grease? And what would help with any barn smell that still lingers on it?

I think it would be more expensive to have it professionally cleaned so it would be best to do it myself, but unsure the best way to clean it.

In order to sanitize the couch and give it a decent cleaning to try to get the odor out. You will most likely have more money wrapped up in a DIY attempt. Then having it professionaly cleaned. DIY "steam cleaners" really do not produce any steam or very hot water. Most are bath temp water. You would want to sanitize the couch and a decent cleaning chem that is safe for the couch would run you a bit too...and then your going to have a ton left over to go to waste.
