I didn't know where to post this, but here goes:

I am planning to put a door into an interior wall where there is currently a larger opening. The floor is a real wood floor, so the question I have is: Should I frame the new wall (surrounding new door frame) right on top of the wood floor (screwing through the wood floor and into the subfloor), or should I cut the space out of the floor for the new bottom plate and screw directly into the subfloor (leaving the finished wood floor to "breathe")?

Thanks for any suggestions.

You can leave the floor in place and install a plate on top of it. If it were carpet, I would probably suggest removing it since it is not a solid product. I am assuming this is full 3/4" hardwood floor and not laminate. In the case of laminate, it must be removed and a 1/4" space left between it and the plate for expansion.

Going on top of a wood floor is fine. If I understand you correctly, your putting a door into this wall your making. If that's the case it's a good idea to make sure your bitting into something solid (not just the wood slats) on the floor and ceiling. :)

Thanks, guys. That seemed the simplest way to do it; I just wanted to make sure I wasn't going to create a problem for the finished floor. Sounds good.
