We mistakenly used a 'Flat' paint in the bathroom. Now it's showing cracks everywhere (not peeling).
Can I simply put a (same color) coat of semi-gloss over the flat paint, or must I put a 'primer' down first?

Thanks.

Flat latex shouldn't crack. What type of paint was under it? What brand/line of flat paint?

As long as the paint is adhered well, there shouldn't be any need for a primer but paint alone probably won't fill or hide the cracks. If they don't sand out, you need to fill them with spackling or joint compound - which would need to be primed.
