For a reason that is nearly impossible to put into words, I need to build a base for a prefabricated toy storage cabinet to sit on. This base would sit, free-standing, on my existing finished floor. It needs to be 7" high, 28" wide, and 14" deep. It would need to support ~100lbs in weight.

This is my first foray into woodworking. I was thinking of making a frame out of 2x6 lumber (actually 5.5" i'm told) sandwiched between two sheets of 3/4" plywood. This would give my the necessary 7" in height.

Would this suffice? Do I just nail everything together, or use some sort of bracket/ connector or construction adhesive?

Your plan will work; however it will not have a "finished" look. The 2x6 frame will be visible and the edges of the 3/4 in. plywood will be exposed. Mitreing the corners when you install the 2x6's and then staining them may be sufficient. The edges of the plywood could be covered with wood ribbon or you could cover with moulding.

Use nails and carpenter's glue.

Having said all the above, I just had another idea. You could use 3/4 inch cabinet grade plywood; rip it 7 inches wide for the sides and ends and cut a piece to fit inside at the top. The top piece would be flush with the top edge of the sides and ends and would be supported by cleats screwed and glued on the inside and 3/4 inch below the top edge. This would stain better than the 2x6's.
