Will a 24 inch pre-hung door assembly fit into a 25 1/2 in.opening? I want to replace the door that is there now, but it is a very odd size.

Only if that 25 1/2" opening is perfectly plumb. The exact size of a new prehung will be very close to 25 1/2". It might be 1/8" smaller. But you usually want a little room around the jamb so as to shim and plumb the frame in the rough opening.
If a doorway is just a little tight, sometimes you can shave one or both sides with a reciprocating saw to make just a little more room for the new door to fit. It helps to check the doorway with a 78" level to find out whether you'll need to shave a little more off the bottom on one side, and maybe the top on the other side. If you do that, you shouldn't have any trouble getting it to fit at all.

Is the 25 1/2" dimension the finish opening or the rough opening.
If it's the rough opening, it might be difficult to adjust a new prehung. It might be easier to just reframe the door opening on one side.
