I'd like to take out my ancient bypass closet doors in one of my bedrooms and install two 24" bi-fold doors. I removed the trim and measured the width of the opening as 49". Height is 82".

Do 24" bi-fold door measurements take into account space required for the jambs or is 49" going to be just a hair too tight for everything?

The finished opening(distance between the side jambs) needs to be 48 inches. 49 inches will work, but you will have wider gaps at the sides and in the center. Good luck with your project.

Is the 49" measurement for the framed opening or is it for the finished opening?

49" is fine. a half inch gap on each side is almost unnoticable. If its only framed then by the time you drywall it will be 48" plus the door you buy is going to be 80" tall. it will look fine
