I want to put up some chair rail molding in the hallway. Is it advisable to nail it up or better to use construction glue? Pros/Cons??

Not the pro around here, but I wouldn't use construction glue alone. Nail it up. ;) Construction glue would be used for better support.
Use spackling compound to the nail holes.

As noted above, constuction adhesive would still need nails to hold it in place until it dries. It will help to better adhere the moulding to the wall but could complicate removal if you change your mind :p
A lot of diyers and some pros  use spackling to fill nail holes but the correct procedure is to use painters putty. If painting, be sure to caulk all the joints to make a professional looking job :D

Ideally, nail into the studs every 16", assuming that is the framing of the house. Angle nails at top/bottom in opposite directions, not straight in. That way they can't pull out. The problem with adhesive is, it squeezes out and has to be cleaned off before it dries.
