I am remodeling a kitchen and want to use a undermount stainless sink that is 18 or 20 inches from frount to back. I have a standard depth countertop (the cabinet is 24" deep) I also want to use the Grohe ladylux Cafe model#33-755 sdo faucet. In the grohe specs it says that I need 4.25 inches between the faucet and the backsplash to allow full movement of the lever. Do I have enough room? I'm not sure?

Probably not. I have an 18" undermount that has 3 1/2" of countertop in front and 3 1/2" countertop in back (plus the backsplash). There is just under 2" between the C/L of the faucet to the backsplash.
Try cutting some cardboard templates and laying it out to see what you get.
BTW - 4 1/2" seems excessive. Have you contacted Grohe to verify the specs?

I need 3.25 inches between faucet and backsplash not 4.25
