I need to replace my single handle kitchen faucet. I have 4 holes in the sink (1 unused, 1 faucet, 1 sprayer, 1 air gap). I would like to replace it with a new single handle with the sprayer pulldown built into the faucet. I show one that I liked that came with a soap dispenser, but I don't think I can use it since I have the Air gap. Is the air gap needed? I assume it is or it would be there. :)

If the air gap (or Air Admittance Valve) was on the old one, then you will need it on the new one. It has to do with the drain venting, not with the water flow. AAV's can terminate under the sink as well, so if routed properly, you can use the extra hole for the soap dispenser. The other three holes will be taken up with the main faucet unit and a cover plate spanning over the other two.
