Can you put down hardi backer on a floor with just screws and no thin-set?

Only if you want your floor to fail.

What should I do if the floor around the tub has a slight squek in it? It seems to be fastened everywhere but I don't want to put the backer down and risk the tiles popping up until I solve this problem.

The squeek could be the wall plate rubbing along it's nails. Try driving a screw at an angle through the bottom plate into the floor. Is this only one layer of ply and how thick?
