I want to increase the strength of my digital cable signal coming into my house.

A. I have a 2 story house. I split my cable at the point just after it comes into my house. I put a signal booster immediately after the split on the side of the split that leads to the second floor. I have a couple of additional splits on that line in my attic. Will it help to add another signal booster somewhere down that line after a split (obviously, it won't help anything prior to the additional signal booster, but will it help after)?

B. Now, going back to the split just after the cable comes into my house...
The cable on this second split goes to a TV less than 5 feet into my house. Will I gain anything by putting a signal booster on this short cable leading to a TV only 3-4 feet away?

I wouldn't boost again. If the TV near the first split is fine I wouldn't amplify it.
If you have RG59 or older RG6, I'd consider replacing it, or at least the main runs.

Thanks for the reply. I have one other question. I saw a booster that is split 4 ways. Is it better to use a couple of splitters down my cable running upstairs as I am already doing (with the single line booster at the start), or use the booster with a 4-way split and run individual cables upstairs?

It is better to home run generally. IMO you would get less los, plus it is flexible down the road.
If you are going to run new wires, run two to each location, for the future.
