I am using 12 inch marble tiles for the backsplash. Currently the wall is covered with a cheap paneling made to look like beadboard. It has a shiney white surface. Can a rough sand the paneling and put the tile right over it or must I use durarock?

Remove that paneling. You can use Durock but regular sheetrock will be ok if the area doesn't get flooded with water spray. For setting marble go with a white modified thinset. Wall mastic may stain the marble.
