I have a Sony DVP-S56OP DVD player that has not had much use. When you put a DVD in, the laser mechanism searches for the disc three times, then says "NO DISC". I've tried several discs and even gone in and cleaned the laser lens. Does anyone have any suggestion for what may be causing this?

It is likely either the laser has failed, or out of alignement, sometimes even a software glitch causes this.
You might try unplugging the unit for 15 minutes, if its just a software glitch, it will reset it to orginal factory setting.

I've unplugged it, but that doesn't help. I had the cover off last night when cleaning the lens and I put a DVD in and tried it. I could see the red light from the lens shining through the DVD, so I would assume that means the laser is functioning. Any other suggestions?

I wish that I did.
You might try MFG see if you can tell you if there has been alot of issues for that model.
Other than that, if its under warranty or parts warranty, then best thing i can suggest is read my helpfull hints and tips.

Problem fixed (at least for now). Did some research on the net and found that it's a common problem with this model. It's the result of a little microfuse inside the unit that ended up being insufficient for what it's used for. The end result is they blow after about a year or so after buying the unit. A guy has a website out there that explains how to remove the fuse and bypass it. The only drawback is it could go again down the road and then I'll have to replace the whole unit. Such is life when you're dealing with companies that care more about the bottom line than actually making a good, reliable product.
