I have an oversize one car garage.20' by 20' I have built a den in the garage 20' by 15' and left a 5' by 20' space for lawn equp and it also has a door to out side in small space which has a little window in it that can be opened for ventilation. I want to put a 36000 btu window unit in the den wall and have the other end sticking in to the small space. I know that it will get hot so I thought I could buy a good fan and exgaust it through the ceiling. and have fresh air come through the door window. Will it work and if it wont can some one give me adv

Yes, it could be done but I don't think it would be practical.
There is a formula for how big the exhaust fan should be to remove the heat from the space.
For a 36,000 btu a/c you would need to exhaust 2250 cfm. When exhausting air you will need to have an opening to allow the air change.
This is a lot of air to move and a pretty big hole.
A better solution would be to make a hole in the wall and mount it directly to the outside.

I hope that 36,000 is for more than that 15x20 den. Otherwise it isn't going to be a den, but a walk-in freezer. A ductless, if you don't have any way to run ducts, with multiple evaporators would be a lot more comfortable & quieter.

36,000 btu is huge for such a small space. That's about enough to cool a 2000 sq ft home.
If the space is insulated pretty good 8,000 - 9,000 btu should be plenty
