When tiling a backsplash, with 3 inch tile and a 3/16 grout line, how much space should there be between the bottom tile and counter top to allow for caulk?

Minimum would be 1/8". You are using a sanded grout and if you want to leave 3/16" at the bottom, go for it. I would suggest you use sanded colored caulk at the line to match the apperance of the rest of the tile.
