We are wanting to put in a new bathroom including toilet and shower into an existing room on a concrete slab. No water is currently in the room. Can holes be drilled into the slab?
What is the best way to go with this project?

First, check with your Building Inspection Department for local codes, permit and inspection requirements.
You're going to have to cut or break up the slab in order to dig trenches for the drain lines to tie it into your existing system.
You also will need to tie the drains into the vent system, and run hot and cold supply lines and your electrical circuits.
The drain lines have to slope 1/4" per linear foot toward the main drain.
Here is a general illustration of an inter-connected drain/waste/vent system:
