I would like to paint over old masonite paneling. How would I go about repairing nail holes? I would liketo us a semi gloss paint. Thank you.

I would try a vinyl spackle to fill and sand flush. Wash the paneling with TSP to ensure that it is clean. Prime with a top quality primer such as Zinsser 123, then paint.
