Hello,
Iam doing framing for a intereior door. After framing king stud, header and jack stud. For putting in a prehung 36'' door what should be the size of the opening. Somebody says 38.5 by 82.5.
Any help would be great.

An inch all the way around should be plenty.

An inch wider that the total width and height of the frame -- not just of the door.
38-1/2" X 82" would work jsut fine.
